In vitro studies showed that Tempol significantly reduced H2O2 mediated mitochondrial respiration and increased LDH release in rat PT cells, indicating a decrease in cell damage and death. The beneficial effects of Tempol are similar to those obtained using Fe2+chelating agent DEF. However, the co administration of DEF and Tempol does not produce any additional beneficial effects against renal ischemia/reperfusion injury or PT cell damage/death mediated by antioxidant stress [2].
In vivo research on SOD simulation Tempol can mimic the effects of resveratrol on cardiac function. Apply Tempol daily through forced feeding method. Compared with oral vectors (VEH), mice treated with Met or Tmp showed reduced PR and QTc intervals, as well as increased heart rate. These results are similar to those obtained through RSV treatment. Cell experiments were conducted to investigate the effects of co administration of Tempol, DEF, and DEF on H2O2 mediated cell damage and death. The confluence culture of PT cells was pre incubated with Tempol (0.03 to 10mM) and DEF (0.03) at 37 ℃ for 10 minutes. DEF (3mM) to 10mM) or DEF (3mM) combined with Tempol (3mM). The concentration range of Tempol and DEF is based on those previously shown in this laboratory to reduce H2O2 mediated cell damage and death (1) cultured rat cardiomyocytes (Tempol) and (2) primary culture of rat PT cells (DEF). Then incubate the PT cell culture with H 2O 2 (1mM) for 4 hours, and then evaluate cell damage and death. After incubation, evaluate cell damage and death using the spectrophotometry described later in this article [2].
