SolifenacinSuccinate, CAS No. 242478-38-2, chemical name 1-azazobicyclic [2.2.2] octane-8-group -(1S) -1-phenyl-3, 4-dihydro-1h-isoquinoline-2-formate succinate, It is a selective muscarinic M3 receptor antagonist developed by Astellas in Japan. It was first marketed in Netherlands, Germany, UK and Denmark in 2004, and in China in 2009. So far, it has been marketed in more than 50 countries and regions around the world. It has become an important drug for the treatment of overactive bladder (OAB) in Europe, the United States and Japan, and has been recommended by several authorities and guidelines. Its preparation is as follows: (S) -1-phenyl-1,2,3, 4-tetrahydroisoquinoline (I) was synthesized by reaction condensation with triphosgene under the condition of organic base (weak base). (S) -1-phenyl-3, 4-dihydro-1h-isoquinoline-2-formyl chloride (II) was obtained. Compound II was heated directly under the catalysis of organic base by one-pot method without separation. It was condensed with (R) -3-quininocyclol to obtain Solinaxine (III), which was then salted with succinic acid to produce Solinaxine (IV) succinate.
